NKX3. 1 is a prostate specific gene encoding a transcription factor that plays an important role in normal prostate development and carcinogenesis. It is a prostatic tumor suppressor gene located on chromosome 8p21. 2, which frequently undergoes a loss of heterozygosity. NKX3. 1 expression is highly restricted in prostate epithelial cells and therefore can be used as a diagnostic biomarker for prostate cancer and other metastatic lesions of prostatic origin. Furthermore, NKX3. 1 shows better sensitivity than Prostate Specific Antigen (PSA) for identifying metastatic prostatic adenocarcinoma. This suggests that immunohistochemical staining of NKX3. 1, along with other prostate-restricted markers, may be valuable for the definitive determination of prostatic origin in poorly differentiated metastatic carcinomas. The NF-E2 DNA binding protein is composed of two subunits, p45 and MafK, and it regulates expression of globin genes in developing erythroid cells through interaction with Maf recognition elements (MAREs). A family of NF-E2 related proteins, which are collectively known as the Cap 'n' collar (CNC) family and include Nrf1 (also designated TCF11), Nrf2 and Nrf3, are bZIP transcription factors that heterodimerize with Maf proteins to bind MARE sequences. The Nrf proteins also bind the antioxidant response element (ARE) and are implicated in the regulation of detoxification enzymes and the oxidative stress response. Recognizes a protein of 75 kDa, identified as mu heavy chain of human immunoglobulins. It does not cross-react with alpha (IgA), gamma (IgG), epsilon (IgE), or delta (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. Monomeric IgM is expressed as a membrane bound antibody on the surface of B cells and as a pentamer when secreted by plasma cells. IgM antibody is prominent in early immune responses to most antigens. Aberrant levels are associated with immune deficiency states, hereditary deficiencies, myeloma, Waldenstrom's macroglobulinemia, chronic infection and hepatocellular disease. Recognizes a protein of 36 kDa, identified as cyclin D1. Cyclin D1, one of the key cell cycle regulators, is a putative proto-oncogene overexpressed in a wide variety of human neoplasms. This antibody neutralizes the activity of cyclin D1 in vivo. About 60% of mantle cell lymphomas (MCL) contain a t(11; 14)(q13; q32) translocation resulting in over-expression of cyclin D1. This antibody is useful in identifying mantle cell lymphomas (cyclin D1 positive) from CLL/SLL and follicular lymphomas (cyclin D1 negative). Stem cell tyrosine kinase (STK-1) has been cloned from a CD34 hematopoietic stem cell enriched library and identified as the human homolog of a previously identified gene of Mouse origin designated either Flk-2 or Flt-3. The STK-1 cDNA encodes a protein of 993 amino acids with 85% identity to Flt-3/Flk-2. STK-1 is a member of the type III receptor tyrosine kinase family that includes Kit (steel factor receptor), Fms and PDGF. STK-1 expression in blood and marrow is restricted to CD34 cells, a population greatly enriched for hematopoietic stem/progenitor cells. STK-1 antiserum recognizes two polypeptides in these cells. The Mouse homolog of STK-1, designated Flt-3/Flk-2, is expressed at high levels in hematopoietic cells and also in neural, gonadal, hepatic and placental tissues. This antibody recognizes a protein doublet of 20-22 kDa, identified as MART-1 (Melanoma Antigen Recognized by T cells 1) or Melan-A. MART-1 is a newly identified melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. Seven other melanoma associated antigens recognized by autologous cytotoxic T cells include MAGE-1, MAGE-3, tyrosinase, gp100, gp75, BAGE-1, and GAGE-1. Subcellular fractionation shows that MART-1 is present in melanosomes and endoplasmic reticulum. This MAb labels melanomas and other tumors showing melanocytic differentiation.
